In need of some help on tuning an amplifier?

I have a digital multimeter, I set it to AC setting and will be using it to tune my gain on my amplifier. I have my head unit turned three quarter way up and my gain all the way down. My amplifier is running at 4 ohms @ 14.4 volts. So I believe I just turn my gain up until the multimeter reaches close to or 14.4 volts? Is that correct? Does it matter that my head unit is putting out 4 volts? Some told me that adjust my gain according to the output on my head unit. Well then if I were to do that my gain would only be at 4 volts.
